Reactive oxygen species (ROS) that are produced by phagocytes or other cells are the main cause of oxidative stress and liver injury. In order to clarify how each of the metabolic factors of liver cells contribute to the liver injury, we studied the acute liver injury caused by massive hepatectomy and the chronic liver injury caused by monocrotaline pyrrole. In the acute liver injury, administration of a nonspecific free radical scavenger, O-2 dimethyl-O-tyrosyl-ribose (2,3,5,6-tetra-O-tyrosyl-beta-D-ribose, TTR), prevented liver injury in the rats subjected to massive hepatectomy. TTR had no effect on the tissue concentration of the ROS in the liver of the rats subjected to massive hepatectomy. Thus, the acute liver injury caused by massive hepatectomy was attributed to the ROS produced by the liver cells themselves. In the chronic liver injury, rats treated with monocrotaline pyrrole showed hepatocyte ballooning and fibrosis in the liver. Both ballooned and non-ballooned hepatocytes were injured by monocrotaline pyrrole, whereas ballooned hepatocytes were more injured than non-ballooned hepatocytes.
